Absurd Trivial Errors in Scatchard Plot Analysis
نویسنده
چکیده
Several types of widespread errors in performing Scatchard plot analysis resulting in fatal distortions of calculated affinities are surveyed more or less systematically. It appears that graphical analysis of non-linear Scathard plot “by eye” is almost impossible; and, moreover, results calculated by computer programs for affinity analysis often require additional "interpretation". Some of reported errors also definitely imply that data fabrication is quite common practice in this field.

Increased affinity of insulin receptor on hepatocytes from streptozotocin-induced diabetic rats.
When we use Scatchard analysis for insulin receptor on intact cells, internalization of insulin may affect the Scatchard analysis. In this study, results from Scatchard analysis for insulin receptor on hepatocytes of streptozotocin (STZ)-induced diabetic rats were compared with those from association and dissociation studies and those from Scatchard analysis using hepatocyte membrane which does...
متن کاملScatchard plot and heterogeneity in binding affinity of labeled and unlabeled ligand.
In saturation analysis the Scatchard plot is a generally accepted method for calculation of the affinity constant, K, and the molar concentration, q, of the binder. However, in a system where the K's for the labeled and unlabeled ligand are unequal, a nonlinear plot can be obtained from which incorrect values for K and q may be calculated. This paper mathematically explains how the plot may dev...
متن کاملSimplified Scatchard-plot assay for estrogen receptor in human breast tumor.
In this two-point Scatchard-plot assay, with which a test of competitive inhibition is combined, the sample is mechanically homogenized in a buffer containing dithiothreitol, ultracentrifuged to obtain a fat-free cytosol, the protein content of which is then adjusted, and free and bound labeled estradiol are separated with dextran-coated charcoal after overnight incubation. We tested the method...
متن کاملThe effect of ligand heterogeneity on the Scatchard plot. Particular relevance to lipoprotein binding analysis.
Computer simulations of equilibrium binding studies of a mixture of two labeled ligands binding competitively to a single class of identical and independent sites (receptors) were performed to investigate how ligand heterogeneity affects the observed data in such studies. The simulated data are presented in Scatchard plots. Ligand heterogeneity was generally found to be indistinguishable from t...
متن کاملUse of mobility ratios to estimate binding constants of ligands to proteins in affinity capillary electrophoresis.
This work evaluates the use of mobility ratios (M) to estimate binding constants of proteins to ligands using affinity capillary electrophoresis (ACE). This concept is demonstrated using two model systems: vancomycin (Van) from Streptomyces orientalis and carbonic anhydrase B (CAB, EC 4.2.1.1). A plot of change in M (deltaM) over the concentration of ligand [L] versus deltaM yields a more usefu...
